Tesamorelin is a 44-amino-acid synthetic analogue of human GHRH. In simple terms, researchers use it to study signaling between the hypothalamic-pituitary axis and the growth hormone system. It acts at the GHRH receptor on pituitary somatotroph cells, activating intracellular pathways that include cyclic AMP and protein kinase A and promoting endogenous growth hormone release.
Unlike compounds that act at the ghrelin receptor, Tesamorelin works through the GHRH receptor and retains the broader regulatory context of pituitary growth hormone secretion. Its N-terminal trans-3-hexenoyl modification was designed to improve resistance to enzymatic degradation compared with native GHRH, making the molecule useful in studies of sustained GHRH-receptor activity.
