Melanotan II, commonly abbreviated MT2 or MT-II, is a synthetic cyclic analog of alpha-melanocyte-stimulating hormone (alpha-MSH). Alpha-MSH belongs to the melanocortin system, a network involved in pigmentation as well as several metabolic and neurological signaling processes. The cyclic structure and inclusion of D-phenylalanine were designed to increase potency and resistance to enzymatic breakdown compared with the naturally occurring hormone fragment.
MT2 acts as a nonselective agonist at several melanocortin receptors. MC1R is closely associated with melanocyte signaling and melanin production, while MC3R and MC4R are frequently investigated in energy-balance, appetite, and neuroendocrine models. MC5R has been studied in exocrine and immune-related signaling. Because MT2 activates multiple receptor subtypes, experimental effects cannot be attributed to a single pathway without receptor-specific controls.
