SLU-PP-332 is a synthetic small-molecule agonist of the estrogen-related receptors ERRalpha, ERRbeta, and ERRgamma, with substantial experimental activity associated with ERRalpha. Despite their name, estrogen-related receptors do not bind estrogen. They are orphan nuclear receptors that help regulate mitochondrial function, oxidative metabolism, energy use, and the expression of genes involved in cellular adaptation.
In cultured skeletal-muscle cells, researchers have reported changes in mitochondrial respiration and the expression of oxidative-metabolism genes. Mouse studies have examined shifts toward more oxidative muscle-fiber characteristics, exercise-responsive genetic programs, energy expenditure, and metabolic markers. These findings led researchers to describe SLU-PP-332 as an experimental “exercise-mimetic” research tool; that term refers only to selected molecular and metabolic patterns observed in controlled preclinical models and does not mean the compound reproduces exercise or its full effects.
