5-Amino-1MQ, also called 5-amino-1-methylquinolinium, is a small-molecule inhibitor of nicotinamide N-methyltransferase. NNMT is an enzyme that converts nicotinamide into 1-methylnicotinamide while using S-adenosylmethionine, or SAM, as a methyl donor. Because nicotinamide is connected to NAD+ salvage and SAM is central to cellular methylation, NNMT can influence several interconnected metabolic pathways.
Cell-based studies have reported that NNMT inhibition can reduce 1-methylnicotinamide, alter intracellular NAD+ and SAM availability, and suppress selected lipogenic processes in adipocytes. Mouse studies have investigated body composition, glucose tolerance, insulin signaling, and adipose-tissue metabolism. These findings are preclinical and do not establish that 5-Amino-1MQ causes weight loss or improves metabolism in humans.
